One reason for the successful spread of the yellow fever mosquito, Aedes aegypti, to at least 17 California counties since 2013--from the Mexican border to Shasta County--may be linked to its resistance to pyrethroids, according to newly published UC Davis research examining genetic markers of resistance at five state locations. 

The work, published in the current edition of Parasites & Vectors, a BioMed Central open-access medical journal, focuses on “determining how informative well-established genetic markers of resistance to pyrethroids are in predicting the resistance phenotype of individual mosquitoes of Aedes aegypti within a population,” said lead author Geoffrey Attardo, medical entomologist-geneticist in the UC Davis Department of Entomology and Nematology. 

“Specifically, we generated mosquito colonies from invasive A. aegypti populations from four locations in the Central Valley (Dinuba, Clovis, Sanger and Kingsburg) and from collections in the Greater Los Angeles Area,”  he said. “Mosquitoes from these populations have all demonstrated resistance to pyrethroid-type insecticides and we think this may be part of the reason why these mosquitoes have been so successful in spreading throughout California.”

 A. aegypti transmits such viruses as dengue, Zika, chikungunya, and yellow fever. Despite California's aggressive surveillance and treatment efforts, this species presents a “significant challenge to local control agencies,” the nine-member team wrote in their research paper, “Frequency of Sodium Channel Genotypes and Association with Pyrethrum Knockdown Time in Populations of Californian Aedes aegypti.“

“What was interesting was that while all the mosquitoes from California show resistance to pyrethroids, there is a lot of variability from one individual to the next in terms of the level of resistance, even when they are carrying genetically identical resistance mutations,” Attardo said. “In particular, there seem to be two levels of resistance in these populations. The two levels seem to represent a resistant group and a super resistant group. However, the proportions of resistant/super-resistant differ in the sampled mosquitoes from population to population.” 

Of particular interest was that mosquitoes carrying the resistance mutations at all five genetic locations were very resistant, he said. “However, there was also a large amount of unexplained variability in terms of the knockdown phenotypes demonstrated by mosquitoes of the same age and rearing conditions. We compared the knockdown times of mosquitoes positive for all five resistance mutations from different populations and found that these mutations account for only a proportion of the observed level of resistance. We believe that the unexplained variability is likely being mediated by the presence or absence of an undefined resistance mechanism.” 

The next step? “Trying to understand what other resistance mechanisms might be present, to identify them, and see if they can be used to develop new methods to monitor pyrethroid resistance levels with a higher degree of accuracy,” Attardo said, adding that “This is especially important as the frequency of the para gene mutations in the populations has increased yearly and will eventually become fixed if this trend continues. This will make them non-informative for use in detection of resistance. If we can develop a more accurate method to measure resistance in mosquito populations, it will be beneficial to vector control agencies/districts for development of pyrethroid resistance management strategies.” 

In launching the project, the researchers designed an assay “to test for the presence of mutations in the gene coding for the pyrethroid target protein, the voltage gated sodium channel (the para gene),” Attardo explained. “Detection of these mutations is used to monitor the level or resistance in populations. However, the actual link between the effect the genotype has on the phenotype of individual mosquitoes has not been looked at in detail. “

The scientists identified mutations from genetic sequences of Californian mosquitoes provided by co-author Yoosook Lee, a former UC Davis mosquito researcher now at the University of Florida-Florida Medical Entomology Laboratory, Vero Beach.

The authors also include research entomologist Anthony Cornel and staff research associate Katherine Brisco of the Mosquito Control Research Laboratory, Kearney Agriculture and Extension Center and UC Davis Department of Entomology and Nematology; and Lindsey Mack, Erin Taylor Kelly, Katherine Brisco, Kaiyuan Victoria Shen, Aamina Zahid, and Tess van Schoor,  all with the UC Davis Department of Entomology and Nematology.

First, they tested the individual resistance phenotype of mosquitoes by placing them into bottles coated with the pyrethroid insecticide permethrin, and observed them to determine how long it takes for them to respond to the insecticide. Said Attardo: “This is a modified version of the assay used by the Center for Disease Control and Prevention to evaluate phenotypic resistance in groups of mosquitoes.” 

Then they isolated the DNA from and performed a high-throughput genetic analysis on each individual to determine the composition of the five mutations in each individual. Next they looked at the resulting data to see how well knockdown time correlates with individual genotypes of mosquitoes. 

Although A. aegypti was first detected in California in 2013, researchers believe that its arrival involved multiple introductions. Populations in Southern California are thought to have crossed the border from Mexico, while Central Valley populations may have been introduced, in part, from the southeastern United States. 

“Upon detection in 2013, the Consolidated Mosquito Abatement District implemented an integrated vector control management strategy which involved extensive public education, thorough property inspections, sanitation, insecticide treatment at larval sources and residual barrier spraying with pyrethroids,” the authors wrote. Despite their efforts, the species successfully overwintered and continued to spread, implicating that it arrived in California with genetic mutations “conferring resistance to the type I pyrethroid insecticides applied for vector control in California.” 

The project drew financial support from the Pacific Southwest Regional Center of Excellence for Vector-Borne Diseases, funded by the U.S. Centers for Disease Control and Prevention.

Medical entomologist Thomas W. Scott, who retired from the UC Davis Department of Entomology and Nematology in 2015 as a distinguished professor but continues his scientific research, is one of 11 UC Davis researchers named to the annual Highly Cited Researchers 2020 list released by the Web of Science Group, the information and technology provider for the global scientific research community. 

The 11 scientists rank in the top 1 percent by citations, which represent how often their papers have been cited in other scientific papers. 

Scott, who joined the UC Davis faculty in 1996, has published 288 papers to date. His total number of citations: more than 33,500. He is internationally known for his work on the ecology and epidemiology of dengue, a mosquito-borne viral infection transmitted mainly by Aedes aegypti.

“Although I retired from UC Davis in 2015, I have continued to carry out research just as I had previously,” Scott said. “In reality, I retired from UC Davis, but I did not retire from science.” 

Scott focuses his research on epidemiology of mosquito-borne diseases, mosquito ecology, evolution of mosquito-pathogen interactions, and evaluation of novel products and strategies for disease control.

“I aim to generate the detailed, difficult to obtain data that are necessary for assessing current recommendations for disease prevention, rigorously testing fundamental assumptions in public health policy, and developing innovative, cost, and operationally effective strategic concepts for prevention of mosquito-borne disease.”

Scott has remained active on a variety of fronts. In 2015, he was in the early stages of two large grants (National Institutes of Health Program Project grant “Quantifying Heterogeneities in Dengue Virus Transmission Dynamics” and a sub-award from a Bill and Melinda Gates grant, “Spatial Repellent Products for Control of Vector-Borne Diseases”), which he continued to run and manage through the Department of Entomology and Nematology. 

“So, I had plenty of support (more than $10 million) to continue my work,” he said. Both projects took place in Iquitos, Peru, and were jointly led with his long-term UC Davis collaborator, Amy C. Morrison, who is in the Department of Pathology, Microbiology and Immunology, School of Veterinary Medicine. 

Scott served as the program leader of the Program Project grant, “which required a lot of work because it had a large cast of international collaborators,” he explained. “That project examined the role of people with mild or unapparent infections in dengue virus transmission dynamics, similar to what is currently being seen with COVID. We determined that people who don't get sick at all or don't get sick enough to go to the doctor can contribute as much as 85 percent to forward transmission; that is,  new infections.”

The Gates grant was a clinical trial to determine whether and to what extent a chemical that repelled mosquitoes would reduce a person's risk of dengue or Zika virus infection. Scott served as the project leader for the Iquitos trial. “Earlier this year we determined that it had a big protective effect,” he related. “We are currently writing that manuscript.”

“Both of those grants ended earlier this year, but we still have a lot of work to do and will be busy for the next couple of years writing papers about those projects,” Scott said. He is currently working as a consultant in a follow-up study on spatial repellents for dengue prevention that will begin in Sri Lanka during 2021.

Since retirement he has been--and continues to be--a member of the Management Committee for  the NGO (non-governmental) Global Dengue and Aedes-Transmitted Diseases Consortium. He co-chairs a Lancet Commission on Aedes-transmitted viruses that is being organized by the NGO.

“Prior to and after retirement, I worked closely with the World Health Organization (WHO), where I served on numerous committees,” Scott related. He chaired the Vector Control Advisory Group, co-chaired the Global Vector Control Response, chaired the Emergency Response Consultation for Zika Virus, chaired the Technical Working Group for Dengue, served on the International Health Regulators Roster of Experts, and co-authored the updated version of WHO's dengue management and control guidelines. He recently applied for membership on the WHO Expert Advisory Group on Arboviruses.

“Working with WHO is important to me because at this stage of my career, being able to translate my science experience into improved quality of life, that is, improved public health policy, for other people, many of whom live in poverty, is the most meaningful thing I can do.” 

Scott, who holds bachelor and master's degrees from Bowling Green (Ohio) State University, received his doctorate in ecology in 1981 from Pennsylvania State University and did postdoctoral research in epidemiology at Yale University School of Medicine's Arbovirus Research Unit, part of the Department of Epidemiology and Public Health. He served on the faculty of the Department of Entomology, University of Maryland, from 1983 to 1996 before joining the UC Davis entomology faculty as a professor of entomology and director of the Vector-Borne Disease Laboratory. He was acting director of the UC Davis Center for Vector-Borne Research from 1996 to 1999, and director of the UC Davis Arbovirus Research Unit (2001-2003). He was selected vice chair of the Department of Entomology in 2006, serving until 2008.

Highly honored by his peers, Scott won the coveted Harry Hoogstraal Medal from the American Society of Tropical Medicine and Hygiene in 2018. His other honors include fellow of three organizations: American Society of Tropical Medicine and Hygiene (2014), Entomological Society of America (2010), and the American Association for the Advancement of Science (2008).  He was named a UC Davis distinguished professor in 2014. In 2015, he won the Charles W. Woodworth Award, the highest honor awarded by the Pacific Branch of the Entomological Society of America. 

Other UC Davis  scientists listed as Highly Cited Researchers include five others from the College of Agricultural and Environmental Sciences: Eduardo Blumwald and Jorge Dubcovsky, both from Plant Sciences; Alan Crozier, Nutrition;  David A. Mills, Food Science and Technology; and Andrew Sih, Environmental Science and Policy.

Doctoral student Olivia Winokur of the UC Davis Department of Entomology and Nematology, is a newly selected fellow of Professors for the Future (PFTF), a program sponsored by UC Davis Graduate Studies “to recognize and develop the leadership skills of outstanding graduate students and postdoctoral scholars who have demonstrated their commitment to professionalism, integrity, and academic service.”

Winokur who enrolled in the entomology doctoral program in 2016 with a designated emphasis in the biology of vector-borne diseases, studies with major advisor Christopher Barker, associate professor, Department of Pathology, Microbiology and Immunology, School of Veterinary Medicine.

As a fellow, she will receive a stipend of $3000. Traditionally, approximately 12 fellows annually are selected to participate in the yearlong program, launched in 1992.

The PFTF program is designed to prepare UC Davis doctoral students and postdoctoral scholars “for an increasingly competitive marketplace and a rapidly changing university environment,” according to PFTF co-directors Ellen Hartigan-O'Connor, acting associate dean of UC Davis Graduate Studies, and Teresa Dillinger, academic administrator.

During the year, the fellows will receive formal training-in-teaching methods and course design; participate in a seminar course on ethics and professionalism, and meet regularly for roundtable panel discussions to promote their professional development, intellectual growth and leadership skills, the directors said.

The fellows will work on projects of their own design to enhance their graduate or postdoctoral experience and professional development of their colleagues. They summarize their projects in end-of-the-year reports.  (See 2019-2020 fellows.)

Winokur titled her successful proposal, “Addressing Financial Barriers to Participation in STEM (Science, Technology, Engineering and Mathematics).”

“Graduate students perform many roles as researchers, mentors, educators, communicators, service leaders, and humans,” wrote Winokur, who is president of the UC Davis Entomology Graduate Student Association. “Financial insecurity affects students' abilities to perform these roles well, and provides a leg up to students with financial support beyond a graduate student stipend. We know that diversity is important in academia; cultivating talent from folks across the social spectrum leads to innovative and appropriate solutions.”

“Addressing financial barriers to participation in STEM graduate programs will lead to more diverse and inclusive programs,” she wrote. “Further, financial insecurity affects those who enters graduate school in the first place; research experience is often required to be considered for acceptance into graduate school, which unfortunately is often offered in the form of unpaid research internships. This can filter out low-income students early, making academia even more elite than it already is.”

Winokur will collaborate with existing resources on campus to set up a series of workshops to address the issues, focusing on three points: (1) creative ways to fund your research (2) how to support your research mentees—why unpaid labor filters low income and other disadvantaged students, and (3) making your teaching cheaper—how to make education more accessible for low-income and other disadvantaged students.

“I've been interested in applying to the Professors for the Future program for a couple years,” she said. “This year, I feel that I am at a good point in my graduate career to develop my skills through the PFTF coursework and to contribute to the graduate student community through a PFTF project.”

Since 2016, the UC Davis doctoral student has developed her teaching, mentoring, course development, and leadership skills through various courses and programs, “which has led to a basic understanding of my teaching philosophy and pedagogy.” She aims to develop her skills to “further align with my core values of diversity, equity, and inclusion in the classroom and laboratory.”

Olivia grew up in Laguna Niguel, Calif. where she focused on science at the Dana Hills High School Health and Medical Occupations Academy. She holds a bachelor's degree, 2015, from Cornell University, where she majored in interdisciplinary studies, focusing on the environmental effects on human health.  While at Cornell, she worked as an intern for the National Institutes of Health, working on climate change initiatives for the National Institute of Environmental Health Sciences and the Fogarty International Center.

Winokur is a National Science Foundation Graduate Research Fellowship. She is a two-time recipient of the Bill Hazeltine Memorial Research Award, given annually to an outstanding UC Davis graduate student studying vector-borne diseases.

Winokur, who researches Aedes aegypti mosquitoes, is the co-author of research published in several journals, including PLOS Neglected Tropical Diseases. Her first first-author paper, “Impact of Temperature on the Extrinsic Incubation Period of Zika Virus in Aedes aegypti” was just published in March. 

Since 2017, she has served as a volunteer with the California Department of Public Health's Vector-Borne Disease Section, assisting with hantavirus and plague surveillance by rodent trapping and testing.

Winokur mentors undergraduate students in the UC Davis Research Scholars Program in Insect Biology (RSPIB), founded and directed by faculty members Jay Rosenheim, Joanna Chiu and Louie Yang, UC Davis Department of Entomology and Nematology.

Active in STEM projects, Winokur co-founded GOALS (Girls' Outdoor Adventure in Leadership and Science) in 2017, a program that develops and runs free two-week summer science programs for high school girls and gender expansive youth from backgrounds underrepresented in STEM fields. The girls learn science, outdoor skills and leadership hands-on while backpacking in Sequoia National Park.      

Meet mosquito researchers Olivia Winokur and Maribel “Mimi” Portilla.

Both are seeking a doctorate at the University of California, Davis. They differ in that Winokur focuses her research primarily on the yellow-fever mosquito, Aedes aegypti, while Portilla's research involves Culex mosquitoes, which transmit West Nile virus and other diseases.

They are alike in that they share the passion of the late William Emery Hazeltine (1926-1994), who worked tirelessly in mosquito research and public health.

Hazeltine, a native of San Jose, was a U.S. Navy veteran who studied entomology at UC Berkeley and received his doctorate in entomology from Purdue University in 1962. He managed the Butte County Mosquito Abatement District, Oroville, from 1966 to 1992, and the Lake County Mosquito Abatement District from 1961-1964.

He was an ardent supporter of the judicious use of public health pesticides to protect public health, remembers Bruce Eldridge, emeritus professor of entomology at UC Davis and former director of the (now folded) statewide UC Mosquito Research Program. Eldridge eulogized him at the 2005 annual meeting of the American Mosquito Control Association (AMCA) as "a man who made a difference." "Bill was a medical entomologist who had a varied career in the field of mosquito biology and control, but he will forever be remembered as a man who fought in the trenches of the pesticide controversy from 1960 until the end of his life, and who made the safe and efficient use of pesticides in public health a personal crusade," Eldridge said.

In his memory, his three sons--Craig Hazeltine of Scottsdale, Ariz., Lee Hazeltine of Lincoln, formerly of Woodland, and the late Jeff Hazeltine (1958-2013)—established the UC Davis Bill Hazeltine Graduate Student Research Awards, awarding the first one in 1997. Each year they travel to Davis to honor the recipients at a luncheon, timed with their attendance at a scholarship and fellowship celebration, hosted by Dean Helene Dillard, UC Davis College of Agricultural and Environmental Science.

Winokur, who studies with advisor Christopher Barker of the Department of Pathology, Microbiology and Immunology, School of Veterinary Medicine, received $2425, and Maribel “Mimi” Portilla, who studies with major professor Sharon Lawler, UC Davis Department of Entomology and Nematology, received $2032.

Winokur's funded project: “Identifying Aedes Mosquito Eggs Using Hyperspectral Imaging: a Rapid, Low-Cost, Non-Destructive Method to Improve Mosquito Surveillance and Control.”

Portilla's project: “The Management of Invasive Weeds and their Effects on Larval Culex mosquitoes.”

“Aedes aegypti and Aedes albopictus are mosquitoes capable of transmitting dengue, chikungunya, yellow fever, and Zika viruses,” Winokur explained in her application. “These species are invasive and present in California and continue to spread, increasing the likelihood of local transmission of these devastating viruses. Additionally, Aedes notoscriptus, an Australian mosquito whose vector competence for many viruses is unknown, has been detected in Los Angeles and is likely to spread in the state.  Aedesmosquitoes are readily detected using ovitraps, a cheap and effective sampling method to detect the presence of gravid females. Ovitraps are especially useful when mosquito populations are low as traps for adult Aedes are unreliable. Once collected, the eggs cannot be differentiated using a stereomicroscope. Traditionally, identifying Aedes eggs collected in ovitraps requires hatching and rearing to adult for visual identification, which is time consuming and leads to a time lag for control, potentially allowing invasive species to spread without intervention.” 

“Currently, I am developing a non-destructive, low-cost method to rapidly identify Aedes eggs,” Winokur wrote. “I have shown that species-specific surface morphologies of the exochorion can be used to differentiate species using electron microscopy. This method is expensive and therefore not a realistic surveillance technique. We can, however, exploit these species-specific surface morphologies in another way to identify Aedeseggs. Slight changes in morphological characteristics can be captured with high spatial resolution proximal sensing imaging, termed hyperspectral imaging.” 

Winokur is testing “the use of  hyperspectral imaging to differentiate between eggs collected from lab colonies of native and invasive Aedes mosquitoes in California. Preliminary data indicate this method shows promise for identifying species and warrants further testing.  Once I have created species-specific reflectance profiles and validated my identification method using colony eggs, I will collect field eggs and validate the identification method using these field eggs.” She is working with  hyperspectral imaging expert Christian Nansen, agricultural entomologist and assistant professor, UC Davis Department of Entomology and Nematology, on the project.

Winokur describes hyperspectral imaging as “a powerful tool that recognizes slight changes; therefore, we need to ensure that all samples are collected and conditioned the same way before testing. Samples must be imaged directly on the oviposition paper because exochorion cells are damaged by the ‘glue' the female uses to attach her eggs to the substrate; imaging removed eggs leads to inconsistent reflectance profiles. This method for rapidly identifying Aedes eggs will allow for quick response to the detection of invasive Aedes mosquitoes.”

 Winokur is using the 2017 Hazeltine funds to improve identification of invasive Aedes mosquito eggs in California and to attend the 2017 American Society of Tropical Medicine and Hygiene conference.  She is also the newly announced recipient of a 2018 Hazeltine Student Research Award for $3,094, this time to investigate Aedes aegypti immune response to Zika virus.

A native of Long Beach, Calif., Olivia grew up in Laguna Niguel, Calif., where she focused on science at the Dana Hills High School Health and Medical Occupations Academy. She received her bachelor's degree in 2015 from Cornell University, majoring in Interdisciplinary Studies and focusing on the environmental effects on human health. She enrolled in the UC Davis graduate program in 2016 as a Ph.D entomology student with a designated emphasis in the biology of vector-borne diseases. Earlier this year, Winokur received a three-year National Science Foundation Graduate Research Fellowship.

After finishing her Ph.D., Winokur plans to remain in academia, but “I'm unsure exactly what that will look like! I really enjoy research, teaching, and mentoring so I'd like to have a career where I can do all of these. I also plan to have a career where I can conduct translational research with broad global health implications, engage non-scientists, create tools to help decision makers mitigate vector-borne disease burden worldwide, and encourage interest and diversity in STEM (science, technology, engineering and mathematics).” 

Maribel Portilla, currently writing her dissertation, said the three chapters will encompass: the management practices of the invasive exotic weeds, Brazilian waterweed (Egeria densa) and water hyacinth (Eichhornia crassipes), in the Sacramento-San Joaquin Delta and how those practices impact mosquitoes and their habitat; herbicide use in managing those weeds and how herbicides affects the larval habitat; and the direct effects of herbicides on larval mosquito development.

Her goal: “to inform and create better techniques to reduce both mosquito and weed problems."

Like Winokur, Portilla is grateful for the Hazeltine research funds. “I am currently exploring some molecular/genetic techniques (PCR and sequencing) to identify the species of mosquitoes I collected in the different weed/habitat mesocosms, This will be funded by the Hazeltine award I received.”

Portilla, who calls the South Bay home (San Martin, Santa Clara County), holds a master's degree in public health from UC Berkeley, where “I was able to study health and disease within a larger context, and learned to consider the biological and the social determinants of disease.” Her area of expertise incorporates her love for biology and her strong interest in social issues.

“At UC Berkeley School of Public Health, I was able to study health and disease within a larger context, and learned to consider the biological and the social determinants of disease. As I completed my degree, I realized I really missed the research experiences I had as an undergraduate. So, I looked for a way to bridge my new-found passion for public health and basic science research. This led me to UC Davis, where I learned about One Health and am now pursuing a Ph.D in medical entomology. Medical entomology is a perfect example of a One Health field, where I can seek out how interactions between humans and animals impact health. I am particularly interested in researching how disease risk may change as people manipulate the environment."

Portilla, who joined the UC Davis Entomology Graduate Program in the fall of 2012, anticipates receiving her doctorate “in six to 12 months.”

Her academic life revolves around writing her dissertation; teaching UC Davis classes (she's taught entomology, general biology and One Health classes); research; and public outreach. Since 2012, she has mentored some 30 undergraduate students on developing and executing their research experiments. She praised the “the diversity of my interns; they each brought such important and unique perspectives to the project.”

What are her career plans?

“Due to my diverse interests and skill set, I am very open about my career choices. I have extensive teaching experience, and would love to be a professor with both teaching and research opportunities. However, there are many opportunities beyond academia. My research is introducing me to many other ways in which my work and research can help keep people safe and healthy. I hope to develop a strong research skill set while at UC Davis, and find a career path which takes advantage of my diverse abilities and love for One Health and Public Health."

Portilla mentioned pursuing a career as a teacher in a small liberal arts school to teach public health, general biology and global diseases classes, as well as do outreach and research. “I'm more of a scientist than an entomologist,” she said.

Portilla may also pursue a career working in vector-control health education at the county, district or state level. “I'm open at this point,” she said. Overall, she is geared toward improving public health outcomes through healthier environments. “I care about how outcomes affect the larger population,” she said.

So did Bill Hazeltine.

“Bill Hazeltine was an advocate for the use of mosquito control to protect people from mosquitoes and the disease agents they transmit, and he believed chemical control to be a necessary part of the means to accomplish this,” wrote Eldridge, who valued his friendship. “He also considered himself an environmentalist, and billed himself as such on his business cards and on his signature block. He had a vast knowledge of pesticides and pesticide legislation, and a strong belief in the scientific basis for public policy issues related to the safe and effective use of pesticides. Because the federal Endangered Species Act influenced mosquito control, he became an authority on this as well."

“After Bill's death, I was contacted by his sons about the possibility of establishing a William Hazeltine Memorial Scholarship Fund at UC Davis,” Eldridge noted. “They believed this would represent an important contribution because of Bill's strong interest and support of medical entomology research, and because of Bill's admiration of UC programs in mosquito control. The fund has grown to the point where graduate student awards can now be funded just from the interest, and a number of students at UCD have benefited from the thoughtfulness of the Hazeltine family.”
